Sensory Neuron-Specific GPCR Mrgprs Are Itch Receptors Mediating Chloroquine-Induced Pruritus<\/p>\n

The cellular and molecular mechanisms mediating
\nhistamine-independent itch in primary sensory
\nneurons are largely unknown. Itch induced by chloroquine
\n(CQ) is a common side effect of this widely
\nused antimalarial drug. Here, we show that Mrgprs,
\na family of G protein-coupled receptors expressed
\nexclusively in peripheral sensory neurons, function
\nas itch receptors. Mice lacking a cluster of Mrgpr
\ngenes display significant deficits in itch induced by
\nCQ but not histamine. CQ directly excites sensory
\nneurons in an Mrgpr-dependent manner. CQ specifically
\nactivates mouse MrgprA3 and human MrgprX1.
\nLoss- and gain-of-function studies demonstrate
\nthat MrgprA3 is required for CQ responsiveness in
\nmice. Furthermore, MrgprA3-expressing neurons
\nrespond to histamine and coexpress gastrin-releasing
\npeptide, a peptide involved in itch sensation,
\nand MrgprC11. Activation of these neurons with the
\nMrgprC11-specific agonist BAM8-22 induces itch in
\nwild-type but not mutant mice. Therefore, Mrgprs
\nmay provide molecular access to itch-selective
\nneurons and constitute novel targets for itch therapeutics.<\/p>\n
