Many studies have addressed how <span class=\"highlight\">TRPC3<\/span> channels are affected by depletion of <span class=\"highlight\">intracellular<\/span> <span class=\"highlight\">calcium<\/span> <span class=\"highlight\">stores<\/span>. Conflicting results have been shown for <span class=\"highlight\">TRPC3<\/span> regarding its function, and this has been linked to its level of expression in various systems. In the present study, we have investigated how <span class=\"highlight\">overexpression<\/span> of <span class=\"highlight\">TRPC3<\/span> interferes with the regulation of <span class=\"highlight\">intracellular<\/span> <span class=\"highlight\">calcium<\/span> <span class=\"highlight\">stores<\/span>. We demonstrate that <span class=\"highlight\">overexpression<\/span> of <span class=\"highlight\">TRPC3<\/span> <span class=\"highlight\">reduces<\/span> the mobilization of <span class=\"highlight\">calcium<\/span> in response to stimulation of the <span class=\"highlight\">cells<\/span> with thapsigargin (TG) or the G-protein coupled receptor agonist sphingosine-1-phosphate (S1P). Our results indicate that this is the result of the expression of <span class=\"highlight\">TRPC3<\/span> channels in the endoplasmic reticulum (ER), thus depleting ER <span class=\"highlight\">calcium<\/span> <span class=\"highlight\">stores<\/span>. OAG evoked <span class=\"highlight\">calcium<\/span> entry in <span class=\"highlight\">cells<\/span> overexpressing <span class=\"highlight\">TRPC3<\/span>, indicating that functional <span class=\"highlight\">TRPC3<\/span> channels were also expressed in the plasma membrane. Taken together, our results show that <span class=\"highlight\">overexpression<\/span> of the putative SOCC, <span class=\"highlight\">TRPC3<\/span>, actually <span class=\"highlight\">reduces<\/span> the <span class=\"highlight\">calcium<\/span> <span class=\"highlight\">content<\/span> of <span class=\"highlight\">intracellular<\/span> <span class=\"highlight\">stores<\/span>, but does not enhance agonist-evoked or store-dependent <span class=\"highlight\">calcium<\/span> entry.
