Here, we show that the neuropeptide natriuretic polypeptide b\u00a0(Nppb) is expressed in a subset of TRPV1 neurons and found that Nppb\u2212\/\u2212 mice selectively lose\u00a0almost all behavioral responses to itch-inducing agents. Nppb triggered potent scratching when\u00a0injected intrathecally in wild-type and Nppb\u2212\/\u2212 mice, showing that this neuropeptide evokes itch\u00a0when released from somatosensory neurons. Itch responses were blocked by toxin-mediated\u00a0ablation of Nppb-receptor\u2013expressing cells, but a second neuropeptide, gastrin-releasing peptide,\u00a0still induced strong responses in the toxin-treated animals.
