However, the possible function of sensory neuron\u00a0<span class=\"highlight\">TLR4<\/span>\u00a0has not yet been formally addressed. In this regard, we investigated the role of\u00a0<span class=\"highlight\">TLR4<\/span>\u00a0in itch signal transmission.<\/p>\n<h4>RESULTS:<\/h4>\n<p><span class=\"highlight\">TLR4<\/span>\u00a0was expressed on a subpopulation of dorsal root ganglia (DRG) sensory neurons that express\u00a0<span class=\"highlight\">TRPV1<\/span>. In\u00a0<span class=\"highlight\">TLR4<\/span>-knockout mice, histamine-induced itch responses were compromised while\u00a0<span class=\"highlight\">TLR4<\/span>\u00a0activation by LPS did not directly elicit an itch response. Histamine-induced intracellular calcium signals and inward currents were comparably reduced in\u00a0<span class=\"highlight\">TLR4<\/span>-deficient sensory neurons. Reduced histamine sensitivity in the<span class=\"highlight\">TLR4<\/span>-deficient neurons was accompanied by a decrease in\u00a0<span class=\"highlight\">TRPV1<\/span>\u00a0<span class=\"highlight\">activity<\/span>. Heterologous expression experiments in HEK293T cells indicated that<span class=\"highlight\">TLR4<\/span>\u00a0expression enhanced capsaicin-induced intracellular calcium signals and inward currents.<\/p>\n<h4>CONCLUSIONS:<\/h4>\n<p>Our data show that\u00a0<span class=\"highlight\">TLR4<\/span>\u00a0on sensory neurons\u00a0<span class=\"highlight\">enhances<\/span>\u00a0histamine-induced itch signal transduction by\u00a0<span class=\"highlight\">potentiating<\/span>\u00a0<span class=\"highlight\">TRPV1<\/span>\u00a0<span class=\"highlight\">activity<\/span>.
